The Isle of Wight Ambulance Service said there are no casualties, but they have sent a car and personnel to be on standby due to the scale of the incident.
A spokesperson for the Hampshire and Isle of Wight Fire and Rescue Service described it is "pretty serious" and said they had sent four pumps, two water carriers and an aerial ladder platform (ALP).
The initial call came in to the emergency services at 7.09pm.
Our reporter at the scene said the thatch is well alight and unfortunately the wind is fanning the flames.
Firefighters are up on the roof (see above photo) in an attempt to control the blaze, and appear to be pulling the thatch off the roof.
The road has been closed from The Sun Inn.
Calbourne is a picturesque village around five miles from Newport.
